The designer - who is famed for her bridal gowns - and her spouse of 23 years have two daughters, Cecilia and Josephine and have vowed to remain amicable for the sake of their children.
A statement released by her company's president, Mario Grauso, said: ''Vera Wang and Arthur Becker have mutually and amicably agreed to separate. They remain devoted parents to their two daughters.''
Becker is believed to have been instrumental in Vera - who was working for Ralph Lauren women's accessories and at-home wear when they married - striking out on her own and advised her as her business developed.
He has also become more involved with the day-to-day operations at Vera Wang in recent years.
A friend of the designer's who asked not to be identified told WWD: ''They will not let this impact the running of the company. They have worked too hard to build it up.''
Along with her successful clothing designs, Vera has also created fragrances and recently launched her own make-up range.
She teamed up with Kohl's stores to release Simply Vera, a line of affordable and stylish cosmetics which includes primers, Shadows, moisturisers and lip colours.
